fludarabine + Vyxeos is a combination chemotherapy regimen being investigated by the University of California, San Diego, for the treatment of newly-diagnosed acute myeloid leukemia (AML). The regimen combines fludarabine, a purine analog antimetabolite, with Vyxeos (CPX-351), which is a liposomal formulation containing cytarabine and daunorubicin in a fixed 5:1 molar ratio. Fludarabine acts by inhibiting DNA polymerase and ribonucleotide reductase, thereby interfering with DNA synthesis. Vyxeos is designed to deliver synergistic concentrations of cytarabine (a pyrimidine analog) and daunorubicin (an anthracycline) directly to the bone marrow, where the liposomes are taken up by leukemia cells. This combination aims to enhance the anti-leukemic activity in patients with intermediate or poor-risk AML by combining the targeted delivery of Vyxeos with the DNA-synthesis-inhibiting properties of fludarabine.
