Overview

This is a combination chemotherapy regimen consisting of four agents: - **Fluorouracil (5-FU):** A pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death in rapidly dividing cells. - **Folinic acid (leucovorin):** Not a cytotoxic agent itself but enhances the binding of 5-FU to thymidylate synthase, increasing its efficacy. - **Mitomycin (commonly as mitomycin C):** An alkylating agent that crosslinks DNA, inhibiting DNA synthesis and function. - **Vincristine:** A vinca alkaloid that inhibits microtubule formation in the mitotic spindle, arresting cell division at metaphase. This combination has been studied primarily for advanced or metastatic gastrointestinal cancers such as colorectal cancer. The regimen leverages multiple mechanisms of action to maximize cytotoxicity against tumor cells. While combinations like 5-FU/folinic acid/mitomycin are established in clinical studies[1][2][3], the addition of vincristine is less common but has precedent in regimens for gastrointestinal malignancies[9]. Each component targets different aspects of cancer cell proliferation and survival.
