fluorouracil + hydroxyurea + paclitaxel is a combination chemotherapy regimen investigated by the GERCOR (Multidisciplinary Oncology Cooperative Group) for the treatment of inoperable stage II or III epidermoid carcinoma of the esophagus. This regimen is administered concurrently with radiation therapy (chemoradiotherapy). It combines the microtubule-stabilizing effects of paclitaxel, the thymidylate synthase inhibition of fluorouracil (5-FU), and the ribonucleotide reductase inhibition of hydroxyurea. These agents act as radiosensitizers, aiming to improve the local control and survival rates of patients with esophageal cancer compared to radiation or dual-agent chemotherapy alone. The regimen was evaluated in a multicenter Phase I/II clinical trial to determine its efficacy and safety profile in this specific patient population.
