Fluorouracil + interferon alfa is a combination regimen of two agents used primarily in oncology. Fluorouracil (also known as 5-FU) is a pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. Interferon alfa is a cytokine with immunomodulatory and antiproliferative effects; it enhances immune responses against tumor cells and can directly inhibit tumor cell proliferation. The combination has been studied for synergistic antitumor activity in several advanced cancers including hepatocellular carcinoma (HCC), colorectal cancer, esophageal cancer, and gastric cancer. Clinical trials have shown objective response rates in these indications with manageable toxicity profiles[1][2][4][6].
