Overview

This is a combination therapy consisting of three agents: - **Fluorouracil (5-FU):** A pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death in rapidly dividing cells. - **Interleukin-2 (IL-2):** A cytokine immunotherapy that stimulates the proliferation and activation of lymphocytes, particularly T-cells and natural killer (NK) cells, enhancing immune-mediated tumor cell killing. - **Mitomycin (commonly as mitomycin C):** An alkylating agent that crosslinks DNA, inhibiting DNA synthesis and function. The combination has been studied for its synergistic antitumor effects. Fluorouracil increases lymphokine activated killer (LAK) activity; IL-2 further boosts immune response; mitomycin’s cytotoxicity is enhanced by immune mediators such as interferon-gamma. This regimen has shown efficacy in clinical studies for hepatic metastases from colon carcinoma when administered via hepatic arterial infusion[1][3][7]. The approach leverages both direct cytotoxicity against tumor cells and stimulation of the host's immune system.
