Overview
FOLFIRI is a chemotherapy combination regimen consisting of three drugs: fluorouracil (also known as 5-fluorouracil or 5FU), irinotecan, and levoleucovorin (the active isomer of folinic acid). Fluorouracil and irinotecan are cytotoxic agents that inhibit DNA synthesis and cell division in rapidly dividing cancer cells. Levoleucovorin enhances the effectiveness of fluorouracil by stabilizing its binding to the target enzyme thymidylate synthase. This combination is primarily used for the treatment of colorectal cancer but may also be used in other cancers as determined by clinical protocols. The regimen works synergistically to maximize antitumor activity while allowing for manageable toxicity profiles[1][2][3][5].
