Overview

Fluoxetine + melatonin is a combination of two agents—fluoxetine, a selective serotonin reuptake inhibitor (SSRI) antidepressant, and melatonin, an endogenous hormone and dietary supplement that regulates circadian rhythms and sleep. This combination has been studied for its potential synergistic effects in alleviating depression-related behavioral abnormalities. Preclinical studies suggest that the combined use of fluoxetine and melatonin may exert greater antidepressant effects than either agent alone, possibly by normalizing brain-derived neurotrophic factor (BDNF)–tropomyosin receptor kinase B (TrkB) signaling in the hippocampus. Clinical data indicate that adding slow-release melatonin to fluoxetine can improve sleep quality in patients with major depressive disorder without significant additional side effects[1][3][5]. However, there are concerns about additive central nervous system depression or sedation when these agents are used together[2][7].
