Fluoxetine + pindolol is a combination therapy consisting of fluoxetine, a selective serotonin reuptake inhibitor (SSRI), and pindolol, a non-selective beta-adrenergic blocker with additional antagonistic activity at serotonin 5-HT1A autoreceptors. This combination has been investigated primarily for the treatment of major depressive disorder and treatment-resistant panic disorder. The rationale behind this combination is that while fluoxetine increases synaptic serotonin by inhibiting its reuptake, its antidepressant effect may be limited by negative feedback from presynaptic 5-HT1A autoreceptors. Pindolol blocks these autoreceptors, potentially enhancing and accelerating the antidepressant response to fluoxetine[1][2][3][6]. Clinical studies have shown that adding pindolol to fluoxetine can increase response rates in depression and improve outcomes in treatment-resistant panic disorder[1][2].
