Overview

Fluoxetine + triiodothyronine is a combination of two pharmaceutical agents used experimentally and off-label to enhance antidepressant efficacy, particularly in treatment-resistant depression. Fluoxetine is a selective serotonin reuptake inhibitor (SSRI) that increases synaptic serotonin levels by inhibiting its reuptake in the presynaptic neuron. Triiodothyronine (T3) is an active thyroid hormone that modulates gene expression and metabolic activity. The combination has been shown to augment and accelerate the antidepressant effects of fluoxetine, potentially through enhanced serotonergic neurotransmission, desensitization of 5-HT1A and 5-HT1B autoreceptors, and increased hippocampal neurogenesis[1][2][7]. This synergistic effect may be especially relevant for patients with major depressive disorder who do not respond adequately to SSRIs alone.
