This combination therapy involves the adjunctive use of flupentixol (brand name Fute) with multi-acting receptor-targeted antipsychotics (MARTAs) for the treatment of schizophrenia. Flupentixol is a thioxanthene-class antipsychotic that acts as a dopamine D1 and D2 receptor antagonist. MARTAs, such as olanzapine and clozapine, are characterized by their affinity for a wide range of receptors, including dopamine D2, serotonin 5-HT2A, histamine H1, and muscarinic receptors. This specific regimen was investigated by Taichung Veterans General Hospital in a Phase 4 clinical trial (NCT04898270) to evaluate whether adding flupentixol could improve clinical efficacy and reduce metabolic syndrome-related side effects, such as weight gain and poor glycemic control, by allowing for a reduction in the MARTA dosage.
