Drug intelligence / Profile preview
**fluzoparib + abiraterone + prednisolone** is an investigational combination regimen being developed by Jiangsu Hengrui Pharmaceuticals for the treatment of prostate cancer, specifically metastatic castration-resistant prostate cancer (mCRPC). The regimen combines fluzoparib (fuzuloparib), an orally active small molecule inhibitor of poly (ADP-ribose) polymerase 1 and 2 (PARP1/2), with abiraterone acetate, an androgen biosynthesis inhibitor targeting CYP17A1, and prednisolone (or prednisone), a synthetic corticosteroid used to prevent mineralocorticoid excess. By simultaneously inhibiting PARP-mediated DNA repair pathways and the androgen receptor signaling pathway, this combination aims to induce synthetic lethality and synergistic antitumor activity in prostate cancer cells. In the Phase III FUZUPRO trial, the addition of fluzoparib to abiraterone and prednisone/prednisolone significantly improved radiographic progression-free survival (rPFS) compared to abiraterone and prednisone/prednisolone alone in patients with mCRPC.
