Overview

Fluzoparib + etoposide is an investigational combination therapy being evaluated for the treatment of recurrent gynecological malignancies, including ovarian, fallopian tube, and primary peritoneal cancers. Fluzoparib is a potent, orally bioavailable small molecule inhibitor of poly (ADP-ribose) polymerase (PARP) 1 and 2, which prevents DNA repair and induces synthetic lethality in cells with homologous recombination deficiencies, such as those with BRCA mutations. Etoposide is a cytotoxic chemotherapy agent that acts as a topoisomerase II inhibitor, causing DNA double-strand breaks by stabilizing the transient complex between the enzyme and DNA. The combination aims to synergistically enhance DNA damage and inhibit repair mechanisms, potentially overcoming resistance to single-agent therapies. This specific regimen is being investigated in Phase 2 clinical trials led by the Liaoning Cancer Hospital and Institute, utilizing fluzoparib developed by Jiangsu Hengrui Pharmaceuticals.
