Overview
FOLFIRI + ramucirumab is a combination regimen used primarily as a second-line treatment for metastatic colorectal cancer (mCRC) and gastroesophageal adenocarcinoma. The regimen consists of the chemotherapy combination FOLFIRI—comprising folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan—administered together with ramucirumab, a monoclonal antibody targeting vascular endothelial growth factor receptor 2 (VEGFR-2). Ramucirumab inhibits angiogenesis by blocking VEGFR-2 signaling, thereby reducing tumor blood supply. The chemotherapeutic agents in FOLFIRI work synergistically to disrupt DNA synthesis and cell division in rapidly proliferating cancer cells. This combination has demonstrated efficacy in prolonging progression-free survival and overall survival in patients whose disease has progressed after first-line therapy[1][3][4].
