FOLFIRINOX + cetuximab is a combination regimen used primarily in the treatment of metastatic colorectal cancer (mCRC), particularly in patients with wild-type KRAS and EGFR-expressing tumors. The regimen combines the chemotherapy protocol FOLFIRINOX—which includes oxaliplatin, irinotecan, leucovorin (folinic acid), and fluorouracil—with the monoclonal antibody cetuximab. Cetuximab targets and inhibits the epidermal growth factor receptor (EGFR), blocking downstream signaling pathways involved in cell proliferation and survival. This dual approach leverages both cytotoxic chemotherapy to kill rapidly dividing cells and targeted therapy to inhibit tumor growth signals[2][4][8]. The addition of cetuximab has been shown to improve efficacy compared to chemotherapy alone in selected patient populations.
