Overview
FOLFIRINOX + panitumumab is an intensive combination therapy regimen consisting of the four-drug chemotherapy backbone FOLFIRINOX (leucovorin, 5-fluorouracil, irinotecan, and oxaliplatin) and the EGFR-targeting monoclonal antibody panitumumab. This regimen was specifically evaluated by the French oncology group UNICANCER in the Phase II PANIRINOX (UCGI28) trial for patients with RAS/BRAF wild-type unresectable metastatic colorectal cancer (mCRC). The treatment strategy utilizes the synergistic effects of multi-agent cytotoxic chemotherapy and targeted EGFR inhibition to maximize tumor shrinkage and potentially convert unresectable metastases to resectable ones. Although the regimen demonstrated high overall response rates, results presented in 2025 indicated that it failed to meet its primary endpoint of complete response in patients with liver-limited disease compared to the standard mFOLFOX6 plus panitumumab regimen.
