FOLFIRINOX3 + bevacizumab is an investigational combination chemotherapy regimen developed by the Centre Georges François Leclerc for the treatment of metastatic colorectal cancer (mCRC). The regimen is a modification of the standard FOLFIRINOX protocol, specifically termed FOLFIRINOX3, which features a desynchronized administration of irinotecan. In this schedule, irinotecan is administered at both the beginning and the end of the chemotherapy cycle (Day 1 and Day 3) to potentially overcome resistance and enhance therapeutic efficacy. This backbone is combined with the anti-angiogenic monoclonal antibody bevacizumab, which targets vascular endothelial growth factor (VEGF). The full regimen includes bevacizumab, oxaliplatin, irinotecan, leucovorin (folinic acid), and 5-fluorouracil. It has been primarily evaluated in Phase I/II clinical trials for patients with mCRC who have progressed after standard first- and second-line treatments.
