FOLFOX + tegafur-uracil is a combination chemotherapy regimen used primarily in the treatment of colorectal cancer. FOLFOX consists of folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin. Tegafur-uracil (UFT) is an oral combination of the prodrug tegafur and uracil; it acts as an alternative oral source of 5-FU with uracil inhibiting dihydropyrimidine dehydrogenase to increase 5-FU bioavailability. The combined regimen targets rapidly dividing tumor cells through multiple mechanisms: - Oxaliplatin forms DNA crosslinks, inhibiting DNA replication and transcription. - Fluorouracil inhibits thymidylate synthase, disrupting DNA synthesis. - Tegafur is converted to 5-FU in the liver, providing sustained cytotoxic activity. This multi-agent approach increases antitumor efficacy by attacking cancer cells at different points in their cell cycle and metabolic pathways[1][2][6][10].
