This research-stage nanomedicine is a pH-responsive targeted drug delivery system designed for the treatment of hepatocellular carcinoma (HCC). It consists of a zinc-based metal-organic framework (Zn-MOF) that serves as a nanocarrier for the chemotherapeutic antimetabolite 5-fluorouracil (5-FU). The MOF core is functionalized with a coating of chitosan conjugated to folic acid (FA-Cs). The folic acid moiety acts as a targeting ligand for folate receptors, which are frequently overexpressed on the surface of cancer cells such as HepG2. The chitosan coating enhances the stability and biocompatibility of the framework. This system is designed to provide controlled release of 5-FU in the acidic microenvironment of tumor cells, thereby increasing therapeutic efficacy while minimizing systemic toxicity to normal cells.
