Folinic acid + interferon-alpha is a combination of two agents, each with distinct mechanisms of action, used primarily in oncology research. Folinic acid (also known as leucovorin) is a reduced form of folic acid that enhances the cytotoxic effects of certain chemotherapeutic agents, notably 5-fluorouracil (5-FU), by stabilizing the binding of 5-FU's active metabolite to thymidylate synthase, thereby increasing inhibition of DNA synthesis[6]. Interferon-alpha is a cytokine and immunomodulator that boosts immune system activity against tumor cells and can directly inhibit cancer cell proliferation through multiple pathways including upregulation of immune effector functions and inhibition of viral replication[7][8]. The combination has been investigated as part of multi-drug regimens for metastatic colorectal cancer and other malignancies, where both drugs act as modulators to enhance the antitumor efficacy—especially when combined with 5-FU[1][3].
