Overview

Fosbretabulin + paclitaxel + carboplatin is a combination regimen investigated primarily for the treatment of anaplastic thyroid cancer (ATC), a rare and aggressive malignancy. Fosbretabulin, also known as combretastatin A-4 phosphate (CA4P), is a vascular-disrupting agent that targets tumor blood vessels by destabilizing microtubules, leading to central necrosis of tumors. It acts as a prodrug and is converted in vivo to its active form, combretastatin A-4[7]. Paclitaxel and carboplatin are established chemotherapeutic agents that inhibit cell division by interfering with microtubule function (paclitaxel) and DNA crosslinking (carboplatin)[2]. The combination has been evaluated in clinical trials for ATC, where it was generally well tolerated but did not show statistically significant improvement in overall survival compared to chemotherapy alone[1][5]. Fosbretabulin was developed by Oxigene.
