Drug intelligence / Profile preview
fosfomycin + trimethoprim-sulfamethoxazole (also known as FTS) is a synergistic combination antibiotic being developed by FleurirABX. It combines fosfomycin, which inhibits the first committed step of peptidoglycan biosynthesis via UDP-N-acetylglucosamine 1-carboxyvinyltransferase (MurA), with trimethoprim-sulfamethoxazole (co-trimoxazole), which inhibits successive steps in bacterial folate biosynthesis via dihydrofolate reductase (DHFR) and dihydropteroate synthase (DHPS). This combination exhibits potent synergistic bacteriostatic and bactericidal activity against a broad spectrum of multidrug-resistant Gram-negative and Gram-positive pathogens, including carbapenem-resistant Enterobacteriales, multidrug-resistant Pseudomonas aeruginosa, carbapenem-colistin-resistant Acinetobacter baumannii, methicillin-resistant Staphylococcus aureus (MRSA), and Enterococcus species. FTS is being developed for potential clinical utility in treating urinary tract infections (UTIs), acute bacterial skin and skin structure infections (ABSSSIs), and drug-resistant respiratory tract infections.
