A combination therapy consisting of fotemustine, a nitrosourea alkylating agent, and vemurafenib, a BRAF inhibitor targeting the BRAF V600E mutation. This combination has been investigated for treating patients with BRAF-mutated metastatic melanoma who have developed resistance to prior BRAF inhibitor monotherapy. Clinical trials have shown that the combination has activity in BRAF-refractory patients with an acceptable safety profile.
