FPA150 + pembrolizumab is a combination investigational immunotherapy regimen composed of FPA150, a monoclonal antibody targeting B7-H4, and pembrolizumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). FPA150 is designed to block B7-H4, an immune checkpoint molecule, potentially enhancing anti-tumor immune responses. Pembrolizumab is an approved PD-1 checkpoint inhibitor that restores T-cell activity against tumors. The combination aims to synergistically enhance immune-mediated anti-cancer effects, particularly in patients with B7-H4 overexpression, and has been studied primarily in B7-H4-positive ovarian cancer. The therapeutic hypothesis is that simultaneous inhibition of B7-H4 and PD-1 can overcome resistance mechanisms in the tumor microenvironment and improve patient outcomes[1][2][3].
