FPI-1434 + pembrolizumab

FPI-1434 + pembrolizumab is an investigational combination therapy being developed by Fusion Pharmaceuticals (a subsidiary of AstraZeneca) in collaboration with Merck. The combination consists of FPI-1434, a targeted alpha radioimmunoconjugate (radiopharmaceutical) targeting the insulin-like growth factor-1 receptor (IGF-1R), and pembrolizumab (KEYTRUDA), a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor monoclonal antibody. FPI-1434 is composed of a humanized monoclonal antibody (AVE1642) targeting IGF-1R, conjugated via Fusion's proprietary Fast-Clear linker technology to the alpha-emitting isotope actinium-225 (225Ac). This combination therapy is designed to target two distinct biological pathways: delivering highly potent alpha-particle radiation directly to IGF-1R-expressing tumor cells while simultaneously blocking the PD-1 pathway to enhance the patient's anti-tumor immune response. The combination is being evaluated in a Phase 1 clinical trial for patients with advanced solid tumors expressing IGF-1R.
