fruquintinib + cetuximab-beta

Fruquintinib + cetuximab-beta is an investigational combination regimen being evaluated for the treatment of RAS wild-type metastatic colorectal cancer (mCRC). Fruquintinib (brand name Elunate/Fruzaqla) is an oral, highly selective small molecule inhibitor of vascular endothelial growth factor receptors (VEGFR-1, -2, and -3), which functions by blocking tumor angiogenesis. Cetuximab-beta is a biosimilar or variant of cetuximab, a chimeric IgG1 monoclonal antibody that targets the epidermal growth factor receptor (EGFR), thereby inhibiting tumor cell proliferation and survival signaling. By combining these two agents, the regimen simultaneously targets the VEGF and EGFR pathways, which are critical for tumor growth and vascularization. This combination is primarily being studied in patients who have failed at least two prior lines of standard therapy (third-line setting) and in first-line settings in combination with immune checkpoint inhibitors.
