fruquintinib + toripalimab

Fruquintinib + toripalimab is a combination therapy consisting of two distinct agents: fruquintinib, an oral, highly selective small molecule inhibitor of vascular endothelial growth factor receptors 1, 2, and 3 (VEGFR1/2/3), and toripalimab, a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1). Fruquintinib inhibits angiogenesis by blocking VEGFR-mediated signaling pathways essential for tumor blood vessel formation. Toripalimab acts as an immune checkpoint inhibitor by binding to PD-1 on T cells, thereby enhancing the immune system’s ability to recognize and destroy cancer cells. This combination has shown synergistic antitumor activity in clinical studies for advanced gastrointestinal cancers such as metastatic gastric or gastroesophageal junction adenocarcinoma and metastatic colorectal cancer after failure of standard therapies[1][2][7].
