FS120 + pembrolizumab is a combination investigational regimen consisting of FS120, a first-in-class tetravalent bispecific antibody that acts as a dual-agonist targeting OX40 and CD137 (also known as 4-1BB), together with pembrolizumab, a programmed death-1 (PD-1)-blocking monoclonal antibody immunotherapy. FS120 binds OX40 and CD137 to stimulate CD4+ and CD8+ T cell activation and proliferation while minimizing hepatotoxicity compared to previous CD137-targeting agents. Pembrolizumab inhibits the interaction between PD-1 and its ligands, resulting in enhanced anti-tumor immune responses. The combination is being studied for synergistic immune activation in oncology, specifically advanced malignancies. FS120 is being developed by F-star Therapeutics; pembrolizumab (Keytruda) is developed and manufactured by Merck.
