FSP77-(AlPcS4A1)6 is a research-stage antibody-drug conjugate (ADC) designed for targeted photodynamic therapy (PDT) of ErbB2-overexpressing cancers. It consists of the internalizing monoclonal antibody FSP77, which specifically targets the ErbB2 (HER2) receptor, covalently conjugated to approximately six molecules of the photosensitizer aluminum tetrasulfonated phthalocyanine (AlPcS4) via a five-carbon sulfonamide spacer chain (A1). The conjugate leverages the receptor-mediated endocytosis of ErbB2 to deliver the photosensitizer intracellularly. Upon exposure to specific wavelengths of light, the photosensitizer generates reactive oxygen species, leading to potent and selective cytotoxicity. In vitro studies on SKOv3 ovarian carcinoma cells have demonstrated significant growth inhibition at very low concentrations, highlighting the efficacy of internalizing antibody-dye conjugates in immunophototherapy compared to non-internalizing alternatives.
