Overview

FT@XBP1 is a nanoparticle-based siRNA delivery system that targets macrophage-specific *Xbp1* deficiency. The formulation uses folic acid-modified D-α-tocopheryl polyethylene glycol 1000 succinate (FA-TPGS) nanoparticles to deliver siRNA against X-box binding protein 1 (XBP1) to hepatic macrophages, thereby inhibiting XBP1 expression. This targeted silencing shifts macrophage polarization from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype, reduces pro-inflammatory factor release, and alleviates hepatic steatosis, liver injury, and fibrosis. Its primary experimental indication is metabolic dysfunction-associated steatohepatitis (MASH)[1].
