Overview

**FT538 + avelumab** is a combination investigational therapy comprising FT538, an engineered induced pluripotent stem cell (iPSC)-derived natural killer (NK) cell product, and avelumab, a PD-L1-blocking monoclonal antibody. FT538 cells are genetically modified to have a high-affinity, non-cleavable CD16 Fc receptor to optimize antibody-dependent cellular cytotoxicity (ADCC), an IL-15/IL-15 receptor fusion to enhance cell persistence, and a CD38 knockout for improved metabolic fitness. Avelumab is a fully human IgG1 monoclonal antibody targeting PD-L1, enabling immune checkpoint blockade and also mediating ADCC via NK cells. This combination is designed to potentiate immune-mediated killing of cancer cells in patients with advanced solid tumors, especially those expressing PD-L1, and has shown preliminary safety and efficacy in phase I clinical trials[1][2][4].
