FT538 + nivolumab is an investigational **combination therapy** consisting of FT538, an off-the-shelf allogeneic natural killer (NK) cell product engineered for enhanced antitumor activity, and nivolumab, a fully human monoclonal antibody targeting the immune checkpoint receptor PD-1 (programmed cell death protein 1). FT538 is genetically engineered to lack CD38 and to express high levels of CD16 and interleukin-15 (IL-15) receptor fusion, increasing NK cell activation and enabling antibody-dependent cellular cytotoxicity (ADCC). Nivolumab blocks PD-1, preventing cancers from evading immune detection and augmenting immune-mediated tumor killing. The combined approach aims to leverage the cytotoxicity of engineered NK cells alongside the immune reinvigoration achieved by checkpoint inhibition. This combination is being evaluated primarily in advanced solid tumors and hematologic malignancies.
