FT538 + trastuzumab is a combination therapy consisting of **FT538**, a multiplexed-engineered, off-the-shelf natural killer (NK) cell therapy derived from clonal induced pluripotent stem cells (iPSCs), and **trastuzumab**, a monoclonal antibody targeting human epidermal growth factor receptor 2 (HER2). FT538 incorporates three synthetic modifications: high-affinity, non-cleavable CD16 Fc receptor for enhanced antibody-dependent cellular cytotoxicity (ADCC) in synergy with monoclonal antibodies; an interleukin-15/IL-15 receptor fusion to promote cell persistence without exogenous cytokine support; and CD38 knockout to improve metabolic fitness and resistance to tumor microenvironment stress. Trastuzumab binds HER2 on tumor cells, triggering immune-mediated cytotoxicity and blocking survival signaling. The combination is being investigated for enhanced anti-tumor activity, particularly in HER2-positive solid tumors, where FT538 enhances trastuzumab-mediated ADCC[1][3].
