Overview

FT576 + daratumumab is a combination investigational therapy under evaluation for relapsed/refractory multiple myeloma. FT576 is an iPSC-derived, allogeneic chimeric antigen receptor (CAR) natural killer (NK) cell product engineered for multi-faceted innate immunity and multi-antigen targeting: it expresses a high-affinity non-cleavable CD16 Fc receptor (hnCD16) to synergize with monoclonal antibodies like daratumumab (enhancing antibody-dependent cellular cytotoxicity, ADCC), an IL-15 receptor fusion (IL-15RF) for cytokine-autonomous persistence, a knockout of CD38 to overcome NK cell fratricide and improve metabolic fitness, and a novel CAR targeting B-cell maturation antigen (BCMA). Daratumumab is an anti-CD38 monoclonal antibody that induces direct and immune-mediated cytotoxicity against CD38+ myeloma cells. The combination is designed to expand anti-myeloma efficacy by targeting both BCMA and CD38 while providing synergistic immune mechanisms—FT576’s engineered NK cells can be activated by daratumumab through their Fc receptor for enhanced killing of myeloma cells[1][5][2][4]. Clinical development is currently in Phase 1 studies[1][3][7][9]. FT576 was developed by Fate Therapeutics and daratumumab (Darzalex) by Janssen/Genmab.
