Fulvestrant + lapatinib is a combination therapy for advanced or metastatic estrogen receptor (ER)-positive breast cancer. Fulvestrant is a selective estrogen receptor degrader (SERD) that binds to and degrades the ER, while lapatinib is a small molecule tyrosine kinase inhibitor targeting both epidermal growth factor receptor (EGFR/HER1) and human epidermal growth factor receptor 2 (HER2/ERBB2). The combination was investigated in clinical trials to determine if adding lapatinib to fulvestrant would improve outcomes in patients with hormone receptor-positive breast cancer, but results showed no significant improvement in progression-free survival or overall survival compared to fulvestrant alone, with increased toxicity associated with the addition of lapatinib[1][6].
