Fulvestrant + ipatasertib is a combination therapy that pairs fulvestrant, an estrogen receptor antagonist, with ipatasertib, an AKT inhibitor. This combination has shown significant efficacy in treating estrogen receptor (ER)-positive, HER2-negative metastatic breast cancer in patients who have progressed on prior aromatase inhibitor and CDK4/6 inhibitor treatment. The combination works by simultaneously blocking estrogen receptor signaling (fulvestrant) and inhibiting the AKT pathway (ipatasertib), which is often activated in resistant breast cancers.
