Overview
Fulvestrant + pirlocyclib is a combination therapy intended for the treatment of hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced or metastatic breast cancer. Fulvestrant is a selective estrogen receptor degrader (SERD) that binds to and accelerates the degradation of estrogen receptors, thereby blocking estrogen signaling in tumor cells. Pirlocyclib is presumed to be a cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, similar in class to palbociclib, ribociclib, and abemaciclib; these agents inhibit CDK4/6 activity and prevent cell cycle progression from G1 to S phase in cancer cells dependent on this pathway for proliferation. The combination aims to provide synergistic antitumor effects by targeting both hormonal signaling and cell cycle regulation.
