Fulvestrant is a selective estrogen receptor degrader (SERD) used in the treatment of hormone receptor-positive (HR+) metastatic breast cancer. This specific regimen, "fulvestrant + placebo," represents the control arm of the Phase II ZAMBONEY trial (NCT00900120) conducted by the Ontario Clinical Oncology Group (OCOG). In this study, postmenopausal women with bone-only or bone-predominant HR+ metastatic breast cancer received 500 mg of fulvestrant via intramuscular injection on a specific schedule (Days 1, 15, 29, and then monthly) combined with a daily oral placebo. Fulvestrant's mechanism involves binding to the estrogen receptor, blocking its action, and inducing its degradation, which effectively downregulates the estrogen signaling pathway necessary for the proliferation of HR+ tumor cells.
