Fulvestrant + pyrotinib is a combination therapy consisting of fulvestrant, a selective estrogen receptor degrader (SERD), and pyrotinib, an irreversible pan-ErbB receptor tyrosine kinase inhibitor. Fulvestrant binds to and accelerates the degradation of the estrogen receptor, thereby inhibiting estrogen signaling in hormone receptor-positive breast cancer cells. Pyrotinib targets multiple members of the ErbB family (including HER1/EGFR, HER2, and HER4), irreversibly inhibiting their kinase activity to block downstream signaling pathways involved in tumor cell proliferation and survival. This combination has been investigated primarily for hormone receptor-positive/HER2-positive metastatic breast cancer patients who have progressed after trastuzumab-based therapies. Clinical studies have shown promising antitumor activity with a favorable safety profile, including robust progression-free survival even in patients with brain metastases[1][2][4].
