Combination therapy consisting of fulvestrant and sapanisertib. Fulvestrant is an estrogen receptor antagonist, while sapanisertib is a selective dual inhibitor of mTOR complexes 1 and 2 (TORC1 and TORC2). This combination was investigated for the treatment of estrogen receptor-positive (ER+)/HER2-negative advanced or metastatic breast cancer in postmenopausal women who experienced disease progression during or after aromatase inhibitor treatment. The combination aims to overcome endocrine resistance by simultaneously targeting the estrogen receptor pathway and the PI3K/AKT/mTOR signaling pathway.
