Futuximab + modotuximab is a fixed 1:1 recombinant human-mouse chimeric IgG1 monoclonal-antibody combination developed as an EGFR-directed antineoplastic therapy. Also known as Sym004, the two antibodies recognize distinct, non-overlapping extracellular EGFR epitopes, producing receptor cross-linking, rapid EGFR internalization and degradation, sustained receptor downregulation, and inhibition of EGFR-driven cancer-cell growth. It has been investigated primarily in anti-EGFR-pretreated, RAS and BRAF wild-type metastatic colorectal cancer, as well as several other EGFR-associated solid tumors.
