Overview

FWD1802 + abemaciclib is an investigational combination therapy being evaluated for the treatment of hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced or metastatic breast cancer. FWD1802 is a novel, orally bioavailable selective estrogen receptor degrader (SERD) developed by Forward Pharma. It acts by binding to the estrogen receptor (ER), inducing its degradation and thereby blocking the primary signaling pathway that drives the growth of ER-positive breast cancer cells. Abemaciclib, developed by Eli Lilly, is a selective small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). By inhibiting these kinases, abemaciclib prevents the phosphorylation of the retinoblastoma protein (Rb), leading to cell cycle arrest in the G1 phase. The combination of a SERD and a CDK4/6 inhibitor is intended to provide a more comprehensive blockade of tumor cell proliferation and overcome potential mechanisms of endocrine resistance.
