Fwd1802 + ribociclib is an investigational combination therapy for estrogen receptor (ER)-positive, human epidermal growth factor receptor 2 (HER2)-negative, unresectable locally advanced or metastatic breast cancer. FWD1802 is a third-generation, oral selective estrogen receptor degrader (SERD) that binds the estrogen receptor (ER) competitively with higher affinity than fulvestrant, aiming to degrade ER and overcome resistance from ESR1 mutations. Ribociclib is an approved, oral small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), designed to suppress cell cycle progression. The combination is being evaluated to maximize anti-tumor activity due to their complementary mechanisms in ER+/HER2- breast cancer, especially for patients who have progressed on previous endocrine therapy.
