Overview

G-GLIP + mitomycin c (also referred to as G-FLIP + mitomycin c) is a multi-agent chemotherapy regimen developed by Hirschfeld Oncology for the treatment of advanced metastatic pancreatic adenocarcinoma. The regimen is a combination of low-dose chemotherapy agents: gemcitabine, fluorouracil (5-FU), leucovorin, irinotecan, and oxaliplatin (collectively known as the G-FLIP regimen), with the addition of mitomycin c. The rationale behind this combination is to utilize lower-than-standard doses of each individual drug to achieve synergistic anti-tumor efficacy while minimizing systemic toxicity and overcoming multi-drug resistance. The components work through diverse mechanisms, including DNA synthesis inhibition, thymidylate synthase inhibition, topoisomerase I inhibition, and DNA alkylation. It is currently being evaluated in a Phase 2 clinical trial for patients with stage IV pancreatic cancer who have progressed after first-line therapy.
