Overview

G250 + DTIn1 is a bispecific monoclonal antibody (bsMAb) produced via quadroma technology, designed for pretargeted imaging and therapy of cancer, notably renal cell carcinoma (RCC)[1][3]. One arm targets the G250 antigen (carbonic anhydrase IX, CAIX) highly expressed on RCC cells, while the other recognizes the DTPA-In hapten via the DTIn-1 monoclonal antibody. In preclinical models, tumors are first pretargeted with the bsMAb, then subsequently targeted with radiolabeled haptens such as ^111^In-DTPA or radioiodinated DTPA-peptides. This strategy enables high tumor-to-background ratios for imaging or therapeutic radionuclide delivery, but is restricted by DTIn-1’s specificity for DTPA chelating indium (not ^90^Y or ^177^Lu).
