GA-TPP + C10 (10-((2,5-dihydroxybenzoyl)oxy)decyl)triphenylphosphonium bromide) is a research-stage small molecule compound designed to target mitochondria in cancer cells. It consists of gentisic acid linked to a triphenylphosphonium (TPP+) cationic group via a ten-carbon aliphatic chain. As a lipophilic cation, it selectively accumulates in the mitochondrial matrix of tumor cells, driven by the mitochondrial membrane potential. Its mechanism of action involves uncoupling the electron transport chain (ETC), inducing metabolic stress, and activating AMPK signaling, which ultimately leads to apoptosis. Preclinical studies have demonstrated its efficacy in models of breast cancer, colorectal cancer, and non-small cell lung carcinoma, particularly when used in combination with agents like doxycycline or 5-fluorouracil. It is currently being investigated by researchers at the University of Chile.
