Galantamine + memantine is a combination of two small molecule drugs used primarily for the treatment of cognitive impairments, most notably in Alzheimer's disease. Galantamine acts as an acetylcholinesterase inhibitor and also serves as a positive allosteric modulator at α4β2 and α7 nicotinic acetylcholine receptors, enhancing cholinergic neurotransmission. Memantine is a noncompetitive antagonist of the N-methyl-D-aspartate (NMDA) receptor, modulating glutamatergic neurotransmission to prevent excitotoxicity. The combination targets both cholinergic and glutamatergic pathways implicated in neurodegenerative diseases, providing synergistic neuroprotective effects. This dual approach aims to address multiple pathological mechanisms underlying cognitive decline[1][2][5].
