Galinpepimut-S + nivolumab is a combination immunotherapy regimen under clinical investigation for the treatment of cancers expressing the Wilms Tumor 1 (WT1) antigen. Galinpepimut-S is a tetravalent peptide cancer vaccine targeting WT1, designed to stimulate both CD4+ and CD8+ T-cell responses against tumor cells overexpressing this antigen. Nivolumab is a monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), thereby enhancing T-cell mediated anti-tumor immunity. The combination aims to potentiate immune responses by both priming tumor-specific T-cells (via galinpepimut-S) and relieving immune suppression in the tumor microenvironment (via PD-1 blockade with nivolumab). This approach has shown promising safety and immunogenicity profiles in early-phase trials for malignant pleural mesothelioma and ovarian cancer[4][7][8].
