Ganaplacide + lumefantrine-SDF is a novel, fixed-dose oral combination therapy under development for the treatment of acute uncomplicated malaria caused by Plasmodium falciparum. Ganaplacide (also known as KAF156) is a new antimalarial agent with an entirely new mechanism of action distinct from current therapies, while lumefantrine is a well-established antimalarial drug here reformulated as a solid dispersion formulation (SDF) optimized for once-daily dosing. This combination aims to address the growing threat of resistance to artemisinin-based therapies and their partner drugs, particularly in Africa where such resistance is spreading. Clinical studies have shown that this regimen is effective and well tolerated in both adults and children, including those infected with artemisinin-resistant parasites. The combination not only clears malaria infection but also has potential to block transmission of the parasite[1][2][4][5][6].
