Overview

Ganetespib + paclitaxel is an investigational combination therapy consisting of ganetespib, a potent small molecule inhibitor of heat shock protein 90 (Hsp90), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Ganetespib disrupts the function of Hsp90, leading to degradation of multiple oncogenic client proteins including mutant p53 and HER2, thereby inhibiting cancer cell growth and survival. Paclitaxel interferes with microtubule dynamics required for mitosis, resulting in cell cycle arrest and apoptosis. The combination has been studied primarily in platinum-resistant ovarian cancer (PROC), HER2-positive metastatic breast cancer, and non-small cell lung cancer (NSCLC). Preclinical studies demonstrated synergistic antitumor activity when both drugs are used together. Clinical trials have shown that this regimen is generally well tolerated at recommended doses (ganetespib 150 mg/m² plus paclitaxel 80 mg/m² weekly) with promising clinical activity in heavily pretreated patient populations[5][6][8][9].
