Overview

**ganitumab + erlotinib** is an experimental combination therapy comprising ganitumab, a fully human monoclonal antibody that targets and inhibits the insulin-like growth factor 1 receptor (IGF1R), and erlotinib, a small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. Ganitumab impedes IGF1R-mediated signaling involved in tumor cell proliferation, survival, and growth, whereas erlotinib blocks EGFR signaling, inhibiting tumor cell proliferation. This combination has been evaluated in phase 1b clinical studies for safety, tolerability, and pharmacokinetics in patients with advanced solid tumors, including ovarian, colon, thyroid, non–small cell lung, squamous cell, and endometrial cancers[1][3]. The rationale for combining these agents is to potentially enhance antitumor efficacy through dual inhibition of the IGF1R and EGFR pathways, which are implicated in tumorigenesis.
